Biological Background: SNW1 Function and Localization
- SNW1 (SNW domain-containing protein 1), also known as Ski-interacting protein (SKIP), Nuclear receptor coactivator NCoA-62, and Bx42, is encoded by the SNW1 gene. It is a multifunctional nuclear protein involved in pre-mRNA splicing and transcriptional regulation.
- As a core component of the spliceosome, SNW1 participates in the splicing of U12-type introns (minor spliceosome) and is required for the specific splicing of CDKN1A pre-mRNA, likely by recruiting U2AF2. - SNW1 modulates transcriptional activity through interactions with numerous factors: it mediates TGF-β signaling via SMAD proteins, MYOD1-dependent transcription via PABPN1, RB1-mediated repression, and RXR/VDR-dependent gene transcription involving coactivators NCOA1 and GRIP1. It also acts as a coactivator for NOTCH1 by binding to multimerized NICD and recruiting MAML1.
