Biological Background: Small Nuclear Ribonucleoprotein F (SNRPF) Function and Localization
- SNRPF, also known as Sm protein F (Sm-F), is a core member of the Sm protein family that assembles into spliceosomal small nuclear ribonucleoproteins (snRNPs).
- It plays a critical role in pre-mRNA splicing as a component of the U1, U2, U4, and U5 snRNPs, which are the building blocks of the major spliceosome. Related references: PMID:11991638, PMID:18984161, PMID:19325628
- SNRPF is found in both the pre-catalytic spliceosome B complex and the catalytically activated C complex, reflecting its dynamic participation in splicing catalysis. Related references: PMID:11991638, PMID:28076346, PMID:28502770
- It also functions in the minor spliceosome, where it mediates the splicing of rare U12-type introns. Related references: PMID:15146077
- In addition, SNRPF participates in histone pre-mRNA 3′-end processing as a subunit of the U7 snRNP. Related references: PMID:12975319
- The protein localizes to both the cytoplasm and nucleus, consistent with its roles in snRNP assembly and splicing.
- Post-translational modifications include acetylation, and the protein is classified as an RNA-binding ribonucleoprotein.
Experimental Guidance and Technical Tips
- For Western blot (WB), a band at approximately 10 kDa is anticipated; due to the small size, use a 15% or gradient gel and appropriate transfer conditions for low-molecular-weight proteins.
- In immunohistochemistry (IHC-P), nuclear staining is expected given SNRPF's subcellular localization; optimize antigen retrieval (e.g., citrate buffer, pH 6.0) and include nuclear markers for co-localization validation.
- The immunogen corresponds to the full-length protein (a.a. 1-86), which may detect both assembled and unassembled forms; consider cross-linking or native gel analyses for snRNP complexes.
- Cross-reactivity with mouse and rat SNRPF is predicted based on sequence homology; however, verify reactivity in the relevant species and tissue type via positive and negative controls.
- For ELISA applications, use recombinant SNRPF or nuclear extract as the coating antigen; assess lot-to-lot consistency for quantitative assays.
