Biological Background: SNRPD1 Function and Localization
- SNRPD1 encodes the small nuclear ribonucleoprotein Sm D1, a core component of the Sm protein family forming the structural scaffold of spliceosomal snRNPs.
- SNRPD1 is a core component of the spliceosomal U1, U2, U4 and U5 small nuclear ribonucleoproteins (snRNPs), essential for pre-mRNA splicing. Related references: PMID:11991638, PMID:18984161, PMID:19325628
- The protein is incorporated into both pre-catalytic B complex and activated C complex spliceosomes, mediating successive steps of the splicing reaction. Related references: PMID:11991638, PMID:26912367, PMID:28076346
- SNRPD1 also functions in the minor spliceosome, facilitating splicing of rare U12-type introns. Related references: PMID:15146077
- The protein may serve as an electrostatic RNA scaffold, promoting snRNP assembly and inter-snRNP contacts via non-specific interactions. Related references: PMID:23333303
- SNRPD1 localizes to both the nucleus and the cytoplasm (including the cytosol), reflecting its roles in snRNP biogenesis and recycling.
- Post-translational modifications include methylation and isopeptide bond formation (Ubl conjugation), which are critical for snRNP assembly and regulatory control.
Experimental Guidance and Technical Tips
- The immunogen comprises the N-terminal 96 amino acids of SNRPD1; consider this region’s conservation when assessing cross-reactivity.
- For Western blot, the predicted molecular weight is ~13 kDa; optimize gel percentage and transfer conditions for small proteins.
- In IHC-P and IF/ICC, validate nuclear and cytoplasmic localization patterns in relevant tissue or cell lines.
- ELISA performance may depend on antigen coating conditions; titrate antibody concentration for optimal signal.
