Biological Background: SNIP1 Function and Localization
- Encoded by the SNIP1 gene in humans, Smad nuclear-interacting protein 1 (SNIP1) is an FHA domain-containing protein that functions as a component of the spliceosome. Required for pre-mRNA splicing, it is specifically involved in the splicing of U12-type introns via the minor spliceosome. Related references: PMID:29360106
- Down-regulates NF-kappa-B signaling by competing with the RELA subunit for binding to the transcriptional coactivators CREBBP/EP300.
- Contributes to the biogenesis of microRNAs (miRNAs), linking splicing and RNA silencing pathways.
- May regulate cyclin-D1/CCND1 mRNA stability through the SNARP complex, which associates with both the CCND1 gene and its mRNA.
- Localizes predominantly to the nucleus, consistent with its roles in splicing and transcriptional regulation.
- Ubiquitously expressed in human tissues, with highest expression levels observed in heart and skeletal muscle.
- Post-translational modifications include phosphorylation and ubiquitin conjugation (isopeptide bond), as identified by proteomics studies.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a central region of human SNIP1 (aa 200–300). For western blot (WB), expect a band at approximately 46 kDa. Validate in your sample system; cross-reactivity with rat has been reported.
- In IHC-P and IF/ICC, nuclear staining is anticipated. Optimize antigen retrieval and permeabilization conditions for your tissue or cell type.
- For ELISA, consider peptide coating or indirect detection methods to assess antibody reactivity.
- Given SNIP1's involvement in multiple pathways, confirm specificity via knockdown or knockout controls where possible.
