Biological Background: SNAP29 Function and Localization
- SNAP29, also known as Synaptosomal-associated protein 29 or Soluble 29 kDa NSF attachment protein, is a SNARE protein essential for cellular membrane fusion.
- As a SNARE, it forms a trans-SNARE complex that drives membrane fusion, and specifically plays a critical role in autophagy by mediating autophagosome-lysosome fusion.
- SNAP29 is also involved in ciliogenesis by regulating membrane fusions necessary for cilium formation.
- The protein localizes to the cytoplasm, Golgi apparatus membrane, autophagosome membrane, cytoplasmic vesicles, and the cilium membrane.
- SNAP29 is widely expressed, found in brain, heart, kidney, liver, lung, placenta, skeletal muscle, spleen, and pancreas.
- It contains a coiled-coil domain and is subject to phosphorylation, as indicated by proteomics identification; it is associated with diseases like ichthyosis, neuropathy, and palmoplantar keratoderma (CEDNIK syndrome).
- Molecular weight is approximately 29 kDa.
Experimental Guidance and Technical Tips
- The recombinant immunogen spans the entire SNAP29 protein (aa 1-258), which may allow recognition of native conformations; consider validating detection of endogenous protein in relevant cell or tissue lysates.
- For Western blot, a band around 29 kDa is expected; post-translational modifications (e.g., phosphorylation) may alter migration; use appropriate controls.
- For ELISA, the antibody can be used for antigen detection; however, optimal working concentrations must be determined empirically.
- Due to cross-reactivity with mouse and rat, this antibody is suitable for studies in these species; confirm specificity in target tissues.
