Biological Background: Zinc finger protein SNAI1 (Snail) Function and Localization

  • Zinc finger protein SNAI1 (Snail homolog 1) is a transcription factor involved in the induction of epithelial-mesenchymal transition (EMT), embryonic mesoderm formation and maintenance, growth arrest, survival, and cell migration.
    Related references: PMID:10655587, PMID:15647282, PMID:20389281
  • Functions as transcriptional repressor by binding to E-boxes in the promoters of target genes (e.g., CDH1, CLDN7, KRT8) and recruiting histone demethylase KDM1A, leading to decreased H3K4 dimethylation.
    Related references: PMID:10655587, PMID:20389281, PMID:20562920
  • The N-terminal SNAG domain competes with histone H3 for binding to the KDM1A/RCOR1 demethylase complex, inhibiting H3K4 demethylation in vitro.
    Related references: PMID:20389281, PMID:21300290, PMID:23721412
  • During EMT, SNAI1 associates with LOXL2 to oxidize histone H3 at pericentromeric heterochromatin, facilitating chromatin reorganization and release of CBX5/HP1A, thereby promoting mesenchymal traits.
    Related references: PMID:16096638
  • Can also activate transcription by cooperating with EGR1 and SP1 to up-regulate CDKN2B in response to TPA, and may independently stimulate the CDKN2B promoter.
    Related references: PMID:20121949
  • Localizes to both the nucleus and cytoplasm.
  • Widely expressed across tissues, with highest levels in kidney; present in mesenchymal and epithelial cell lines.
  • Classified as a zinc-finger protein with key roles in development, DNA binding, and multiple post-translational modifications (ADP-ribosylation, phosphorylation, ubiquitination).

Experimental Guidance and Technical Tips

  • The immunogen spans amino acids 1-264 of human SNAI1, encompassing the full-length protein including the N-terminal SNAG domain and the zinc-finger regions, which may contribute to broad epitope recognition.
  • For western blotting, a band around 29 kDa is expected; consider including nuclear and cytoplasmic fractionation controls given its dual localization.
  • In IHC-P and IF/ICC experiments, primary localization is expected to be nuclear, but cytoplasmic staining may also be observed depending on cell state and EMT induction.
  • Cross-reactivity with mouse and rat SNAI1 is indicated; validate specificity in your experimental system using appropriate controls.
  • This polyclonal antibody may detect multiple isoforms or post-translationally modified forms; for critical applications, consider confirming results with orthogonal methods.
