Biological Background: SMYD5 Function and Localization
- SMYD5 (SET and MYND domain-containing protein 5), also known as RRG1, RAI15, NN8-4AG, and ZMYND23, is a protein-lysine N-trimethyltransferase belonging to the SMYD family.
- It specifically trimethylates Lys-22 of the RPL40/eL40 ribosomal subunit, promoting translation elongation and protein synthesis. Related references: PMID:39048817 PMID:39103523
- SMYD5 may also function as a histone methyltransferase, catalyzing trimethylation of histone H3 at Lys-36 (H3K36me3) and histone H4 at Lys-20 (H4K20me3) in the context of histone octamers, though its activity on nucleosomes is uncertain in vivo. Related references: PMID:39048817 PMID:39103523
- In macrophages, SMYD5 associates with the NCoR corepressor complex to repress toll-like receptor 4 (TLR4)-target inflammatory genes, likely through deposition of H4K20me3 marks at gene promoters.
- It plays a critical role in embryonic stem (ES) cell self-renewal and differentiation by maintaining genome stability through heterochromatin formation and silencing of endogenous repetitive DNA elements via H4K20me3. Related references: PMID:28951459
- SMYD5 is also involved in the hypothermia response: mild hypothermia triggers its degradation, relieving H3K36me3-mediated repression at the SP1 neuroprotective gene promoter.
