Biological Background: SMYD3 Function and Localization

  • SMYD3 (SET and MYND domain-containing protein 3) is a histone-lysine N-methyltransferase that specifically methylates histone H3 at lysine 4 (H3K4me2/me3) and histone H4 at lysine 5 (H4K5me). Related references: PMID:15235609 PMID:22419068
  • It functions as a transcriptional co-activator by binding DNA sequences 5'-CCCTCC-3' or 5'-GAGGGG-3' and associating with RNA polymerase II complexes. Related references: PMID:15235609
  • The protein is also known as KMT3E, ZMYND1, and ZNFN3A1, and contains a SET domain for methyltransferase activity and a MYND-type zinc finger domain.

  • This antibody has been tested in Western blot (WB), immunofluorescence/immunocytochemistry (IF/ICC), and ELISA. For WB, a band at ~49 kDa is expected. Appropriate loading controls and blocking conditions should be optimized.
  • Cross-reactivity with mouse and rat SMYD3 is reported; however, sequence alignment of the immunogen region should be confirmed in the relevant species to ensure specificity.
