- Methyltransferase activity on p53/TP53 is higher than on histones; SMYD2 monomethylates p53 at Lys-370, reducing its DNA-binding and transcriptional activity.
- SMYD2 also monomethylates RB1 at Lys-860, potentially regulating tumor suppressor functions.
- The methyltransferase activity of SMYD2 requires interaction with HSP90alpha.
- SMYD2 contains a SET domain and a MYND-type zinc finger, classifying it as a member of the SMYD family of methyltransferases.
