Biological Background: Myosin-11 (SMMHC/MYH11) Function and Localization

  • Myosin-11, also known as smooth muscle myosin heavy chain (SMMHC), is a motor protein that powers muscle contraction via actin binding and ATP hydrolysis.
  • Encoded by the MYH11 gene, alternative splicing generates isoforms, including isoform 1 prevalent in slowly contracting tonic smooth muscles.
  • It localizes to the thick filament of smooth muscle myofibrils and has been detected in melanosomes, suggesting extra-contractile roles.
  • Predominantly expressed in smooth muscle tissues such as the umbilical artery, bladder, esophagus, and trachea.
  • Post-translational modifications, including phosphorylation and methylation, regulate myosin-11 assembly, activity, and disease associations.
  • Mutations and chromosomal rearrangements involving MYH11 are linked to aortic aneurysm and vascular pathologies, and it is recognized as a proto-oncogene in certain contexts.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 1160–1313; consider this region when designing peptide competition assays or epitope mapping.
  • For Western blot, validate the antibody with lysates from smooth muscle cell lines or tissues (human, mouse, rat). Note the high molecular weight (~227 kDa); use low-percentage SDS-PAGE gels and extended transfer times.
  • In ELISA, optimize coating antigen concentration and antibody dilution using recombinant protein or peptide. Cross-reactivity with other myosin heavy chains should be assessed empirically.
  • This antibody has not been tested in IHC or IF; if required, perform pilot experiments with appropriate fixation and permeabilization protocols.
