Biological Background: SMCHD1 Function and Localization
- SMCHD1 (Structural maintenance of chromosomes flexible hinge domain-containing protein 1), also known as BAMS or FSHD2, is a non-canonical member of the structural maintenance of chromosomes (SMC) protein family.
- As a chromatin regulator, SMCHD1 promotes heterochromatin formation on both autosomes and the X chromosome, probably by mediating the merge of chromatin compartments.
- It plays a key role in chromosome X inactivation in females by promoting the spreading of heterochromatin; it is recruited by Xist RNA and facilitates Xist RNA spreading across the inactivated X chromosome. Related references: PMID:23542155
- SMCHD1 is required for silencing of a subset of clustered autosomal loci in somatic cells, such as the DUX4 locus—a function linked to facioscapulohumeral muscular dystrophy type 2 (FSHD2). Related references: PMID:23143600
- The protein possesses ATPase activity and may participate in ATP‑dependent structural manipulation of chromatin to regulate gene expression. Related references: PMID:29748383
- SMCHD1 contributes to DNA repair: it localizes to sites of DNA double‑strand breaks and promotes repair via non‑homologous end joining (NHEJ) while inhibiting homologous recombination (HR). Related references: PMID:24790221 PMID:25294876
- Subcellularly, SMCHD1 is localized to chromosomes, consistent with its functions in chromatin organization and DNA repair.
- Post‑translational modifications indicated by UniProt keywords include acetylation, phosphorylation, and ubiquitin conjugation; alternative splicing may generate multiple isoforms.
Experimental Guidance and Technical Tips
- The immunogen targets amino acids 1756‑2005 in the C‑terminal region; consider potential isoform‑specific detection and verify reactivity with your target sequence.
- For Western blot, expect a band at approximately 226 kDa. Use high‑molecular‑weight markers and optimize transfer conditions for large proteins.
- In immunofluorescence/immunocytochemistry (IF/ICC), SMCHD1 should localize to chromatin‑rich regions; include appropriate controls for nuclear staining.
- ELISA applications may be developed using recombinant SMCHD1 or cell lysates; validate linearity and specificity with your sample set.
- As a rabbit polyclonal antibody, monitor inter‑lot consistency if used in quantitative or regulated assays.
- Cross‑reactivity with rat is predicted based on sequence similarity; confirm performance in your model system.
