• SMC5 (Structural maintenance of chromosomes protein 5), also known as SMC5L1 and ATELS2, is a core component of the SMC5-SMC6 complex essential for DNA double-strand break repair by homologous recombination.
• The complex promotes sister chromatid homologous recombination by recruiting the SMC1-SMC3 cohesin complex to double-strand breaks.
• Required for telomere maintenance via recombination in alternative lengthening of telomeres (ALT) cell lines and mediates sumoylation of shelterin complex components, leading to shelterin disassembly in ALT-associated PML bodies (APBs).
