Biological Background: SMARCC2 Function and Localization
- SMARCC2, also known as BAF170, is a core subunit of the SWI/SNF chromatin remodeling complex, belonging to the SMARCC family and found in both canonical BAF and PBAF complexes.
- It is involved in transcriptional activation and repression by altering DNA-nucleosome topology in an ATP-dependent manner (PubMed:11018012). It can stimulate the ATPase activity of the catalytic subunit of these complexes (PubMed:10078207). Related references: PMID:11018012 PMID:10078207
- SMARCC2 may be required for CoREST-dependent repression of neuronal gene promoters in non-neuronal cells (PubMed:12192000). Related references: PMID:12192000
- It is a component of both the neural progenitors-specific (npBAF) and neuron-specific (nBAF) chromatin remodeling complexes, which undergo subunit switching during neural differentiation to regulate stem cell self-renewal and dendrite growth, respectively (By similarity).
- SMARCC2 is a critical regulator of myeloid differentiation, controlling granulocytopoiesis and neutrophil granule formation gene expression (By similarity).
- Localizes exclusively to the nucleus, and is ubiquitously expressed. Post-translational modifications include acetylation, phosphorylation, ADP-ribosylation, and ubiquitination, while disease-associated variants are linked to intellectual disability.

- The immunogen is a synthetic peptide spanning amino acids 150–250 of human SMARCC2. Align this region with mouse and rat sequences to confirm cross-reactivity expectations.
- For Western blotting, SMARCC2 has a predicted molecular weight of ~133 kDa. Use a low-percentage acrylamide gel (e.g., 8%) and extended transfer times for efficient high-molecular-weight protein transfer.
- When performing ELISA, optimize antibody and antigen concentrations via checkerboard titration. Include appropriate positive (e.g., recombinant SMARCC2) and negative controls to verify specificity.
