Biological Background: SMARCC1/BAF155 Function and Localization
- SMARCC1 (SWI/SNF-related matrix-associated actin-dependent regulator of chromatin subfamily C member 1), also known as BAF155, BRG1-associated factor 155, is a core subunit of the SWI/SNF chromatin remodeling complexes.
- It plays a critical role in transcriptional activation and repression by altering DNA-histone contacts in an ATP-dependent manner and may stimulate the ATPase activity of the catalytic subunit. Related references: PMID:10078207 PMID:29374058
- SMARCC1 is a component of neural progenitor-specific (npBAF) and neuron-specific (nBAF) complexes, where subunit switching during differentiation is essential for neural stem cell self-renewal and dendrite growth.
- The protein localizes to both the nucleus and cytoplasm.
- It is broadly expressed in tissues including brain, heart, muscle, placenta, lung, liver, kidney, and pancreas.
- SMARCC1 is subject to multiple post-translational modifications such as acetylation, methylation, phosphorylation, and ubiquitination, and is associated with neurogenesis and disease variants.
Experimental Guidance and Technical Tips
- This antibody was generated using a recombinant protein corresponding to amino acids 700-800 of human SMARCC1/BAF155, which may provide specific recognition of this region.
- In Western blot (WB), a predominant band at ~123 kDa is expected; consider using appropriate loading controls and validate in your sample system.
- For immunofluorescence (IF/ICC), anticipate nuclear and potentially cytoplasmic staining; fixation and permeabilization conditions should be optimized.
- Chromatin immunoprecipitation (ChIP) may be feasible; however, due to the large size of the SWI/SNF complex, it may require careful optimization of crosslinking and shearing protocols.
- Cross-reactivity is reported for human and mouse; for other species, sequence homology should be checked.
