Biological Background: SMARCAD1 Function and Localization

• SMARCAD1 (SWI/SNF-related matrix-associated actin-dependent regulator of chromatin subfamily A containing DEAD/H box 1) possesses intrinsic ATP-dependent nucleosome-remodeling activity and is required for both DNA repair and heterochromatin organization. Experimental Guidance and Technical Tips

• Based on the immunogen sequence (amino acids 928–1026), this antibody recognizes the C-terminal region of SMARCAD1. For Western blot (WB), prepare lysates using standard RIPA buffer and probe for bands near 117 kDa and a smaller isoform at ~68 kDa; include positive controls such as fibroblast or keratinocyte extracts.
• For immunoprecipitation (IP), use 1–2 µg of antibody per 500 µg of cell lysate and validate co-precipitation with known interaction partners or negative IgG controls.
• In immunofluorescence (IF/ICC) and immunohistochemistry (IHC-P), fix cells or tissue sections with 4% paraformaldehyde and permeabilize with 0.1% Triton X-100; nuclear staining patterns are expected. Optimize antigen retrieval for paraffin sections.
• For ELISA applications, coat plates with recombinant SMARCAD1 fragment or cell/tissue lysates and perform a checkerboard titration to determine optimal antibody and antigen concentrations.
• Given cross-reactivity with human, mouse, and rat, the antibody is suitable for multi-species studies; however, verify performance in your specific experimental system.
