Biological Background: SMARCA5/SNF2H Function and Localization
- SMARCA5, also known as SNF2H or hSNF2H, is a SWI/SNF-related matrix-associated actin-dependent regulator of chromatin subfamily A member 5.
- It functions as an ATP-dependent nucleosome-remodeling factor, serving as the catalytic subunit of ISWI chromatin-remodeling complexes that facilitate DNA replication, transcription, and repair (PubMed:12972596, PubMed:28801535, PubMed:10880450, PubMed:12198550, PubMed:12434153). Related references: PMID:12972596 PMID:28801535 PMID:10880450 PMID:12198550 PMID:12434153
- SMARCA5-containing ISWI complexes slide histone octamers along DNA, and full remodeling activity may require intact histone H4 tails (PubMed:10880450, PubMed:28801535). Related references: PMID:10880450 PMID:28801535
- It is a key component of several chromatin-remodeling complexes, including ACF, WICH, NoRC, and RSF, each exhibiting distinct nucleosome spacing and sliding properties (PubMed:14759371, PubMed:15543136, PubMed:28801535). Related references: PMID:14759371 PMID:15543136 PMID:28801535
- SMARCA5 localizes to the nucleus and is associated with chromosomes, consistent with its role in maintaining chromatin structure.
- It is ubiquitously expressed across tissues, reflecting its fundamental role in chromatin dynamics.
- Post-translational modifications include acetylation, phosphorylation, and ubiquitin conjugation, which may regulate its activity in response to cellular signals.
- Additionally, SMARCA5 is involved in host-virus interactions and is essential for replication of pericentric heterochromatin (PubMed:12434153). Related references: PMID:12434153
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal ATPase/helicase domain (aa 100-400) of human SMARCA5; consider using this antibody for Western blot (WB) and ELISA applications.
- For WB, the predicted molecular weight of SMARCA5 is ~122 kDa; optimize gel percentage and transfer conditions accordingly.
- Validate antibody reactivity in your specific sample system; SMARCA5 is ubiquitously expressed, so a wide range of cell lines and tissue lysates may be suitable.
- Cross-reactivity with mouse SMARCA5 has been reported; this enables comparative studies between human and mouse models.
