Biological Background: SMARCA5/SNF2H Function and Localization
- SMARCA5, also known as SNF2H or ISWI, is the catalytic subunit of ISWI chromatin-remodeling complexes with intrinsic ATP-dependent nucleosome-remodeling activity. Related references: PMID:12972596, PMID:28801535
- It forms complexes with BAZ1A/ACF1, BAZ1B/WSTF, BAZ2A/TIP5, BAZ2B, CECR2, and RSF1 to regulate nucleosome spacing and sliding, and is essential for assembling regular nucleosome arrays. Related references: PMID:12972596, PMID:14759371, PMID:15543136, PMID:28801535
- SMARCA5 is required for DNA replication of pericentric heterochromatin in S-phase, in conjunction with BAZ1A/ACF1. Related references: PMID:12434153
- It participates in RNA polymerase I and III transcription through the WICH and B-WICH ISWI chromatin-remodeling complexes. Related references: PMID:11980720, PMID:15543136, PMID:16603771
- Subcellularly, SMARCA5 localizes to the nucleus and chromosomes.
- Ubiquitously expressed, it undergoes post-translational modifications including acetylation, phosphorylation, and ubiquitination.
- SMARCA5 mediates histone H2AX phosphorylation at Tyr-142 and maintains chromatin structure during DNA replication.
- Additional associated keywords: 3D-structure, host-virus interaction, isopeptide bond, repeat, reference proteome.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide from the C-terminal region (aa 953-1052); consider this for epitope specificity in applications.
- For Western blot (WB), detect SMARCA5 at approximately 122 kDa; validate with appropriate positive controls.
- In immunofluorescence (IF/ICC), nuclear permeabilization is recommended due to its chromatin localization.
- For ELISA, optimize coating and detection conditions based on the target antigen.
- Cross-reactivity with mouse and rat allows for versatile model organism studies; validate in the relevant species tissue or cell line.
- Pair with appropriate secondary antibodies and detection reagents for consistent results.
