Biological Background: Smad5 Function and Localization
- SMAD5 is the product of the SMAD5 (MADH5) gene and is also known as JV5-1, DWFC, and Mothers against decapentaplegic homolog 5.
- SMAD5 functions as a transcriptional regulator in the BMP signaling pathway, playing critical roles in embryonic development, cell differentiation, angiogenesis, and tissue homeostasis (PubMed:12064918, PubMed:16516194). Related references: PMID:12064918, PMID:16516194
- Upon BMP receptor activation, SMAD5 is phosphorylated, associates with SMAD4, and translocates into the nucleus where the complex recognizes Smad Binding Elements (SBEs) to regulate gene expression (PubMed:9442019, PubMed:33510867). Related references: PMID:9442019, PMID:33510867
- Non-phosphorylated SMAD5 resides in the cytoplasm and mitochondria, where it regulates energy metabolism by promoting mitochondrial respiration and glycolysis via interaction with hexokinase 1 (HK1) (PubMed:28675158). Related references: PMID:28675158
- SMAD5 is a member of the receptor-regulated SMAD (R-SMAD) family that mediates signals from the TGF-β superfamily.
- It is ubiquitously expressed across tissues and undergoes post-translational modifications including phosphorylation and acetylation, which modulate its activity.
- Subcellular localization is dynamic: predominantly cytoplasmic in the basal state, nuclear upon activation, and also detected at mitochondria.
Experimental Guidance and Technical Tips
- For Western blotting, expect a specific band near 52 kDa corresponding to SMAD5; validate in the appropriate cell or tissue lysate (human, mouse, or rat origin).
- This antibody is suitable for ELISA applications; optimal working dilutions should be determined empirically for each assay format.
- The immunogen (aa 186-260) targets a conserved region; while cross-reactivity with closely related SMADs is possible, this antibody has been validated for recognition of human, mouse, and rat SMAD5.
