Biological Background: SLFN11 Function and Localization

  • SLFN11 is a member of the Schlafen family that functions as a guardian of the genome by inhibiting DNA replication and promoting cell death in response to DNA damage. It persistently blocks stressed replication forks by opening chromatin at initiation sites, possibly unwinding DNA ahead of the MCM helicase (PubMed:29395061, PubMed:22927417, PubMed:26658330).   • Upon DNA damage, SLFN11 inhibits translation of ATR or ATM based on distinct codon usage without disrupting early DNA damage response signaling (PubMed:30374083). Related references: PMID:30374083
