Biological Background: SLCO1B1 Function and Localization
- SLCO1B1 (Solute carrier organic anion transporter family member 1B1), also known as OATP-C, LST-1, OATP2, OATPC, and SLC21A6, is a liver-specific transmembrane transporter that mediates Na⁺-independent uptake of a wide range of organic anions, including bile acids, conjugated steroids, eicosanoids, and thyroid hormones. Related references: PMID:10358072, PMID:15159445, PMID:17412826
- SLCO1B1 contributes to the hepatic detoxification loop by transporting bilirubin glucuronides from plasma into hepatocytes, facilitating their clearance and reducing systemic toxicity. Related references: PMID:22232210
- This transporter plays a critical role in the pharmacokinetics of clinically important drugs, including HMG-CoA reductase inhibitors (statins like pravastatin and pitavastatin), the chemotherapeutic methotrexate, and antihypertensive agents such as enalapril and valsartan. Related references: PMID:10601278, PMID:15159445, PMID:15970799, PMID:23243220, PMID:16624871, PMID:16627748
- SLCO1B1 also transports coproporphyrin I and III, by-products of heme synthesis, suggesting a role in hepatic porphyrin disposition. Related references: PMID:26383540
- Substrate transport by SLCO1B1 is pH-sensitive, with enhanced activity in acidic microenvironments, and likely involves HCO₃⁻ as the counteranion exchanged for organic anions. Related references: PMID:19129463
- The protein localizes to the basolateral membrane of hepatocytes, enabling efficient uptake of substrates from sinusoidal blood. It is highly expressed in the liver, particularly in centrilobular hepatocytes, with low or absent expression in most other tissues. Related references: PMID:10358072, PMID:10601278, PMID:10873595, PMID:12196548, PMID:22232210
- In the testis, SLCO1B1 is detected at the basal membrane of Sertoli cells and weakly in Leydig cells, indicating a potential role in the blood-testis barrier. Related references: PMID:35307651
- Post-translational modifications include glycosylation and phosphorylation, and the protein contains disulfide bonds. It belongs to the major facilitator superfamily of transporters.
Experimental Guidance and Technical Tips
- The immunogen (amino acids 597–691 of human SLCO1B1) corresponds to a C-terminal intracellular region, which is well conserved across species. This polyclonal antibody is suitable for detecting endogenous SLCO1B1 in human, mouse, and rat samples by WB and ELISA.
- For Western blotting, the predicted molecular weight is approximately 76 kDa; consider using liver tissue lysates or hepatocyte-derived cell lines as positive controls, given the high hepatic expression.
- Due to the broad substrate specificity of SLCO1B1, functional studies may require careful control of endogenous organic anion levels. The antibody can be used to monitor SLCO1B1 expression changes under different pharmacological or pathological conditions.
- As a polyclonal antibody, lot-to-lot variability may occur; pilot experiments are recommended to determine optimal working dilutions for each application and sample type.
