Biological Background: SLC7A11/xCT Function and Localization

  • SLC7A11 (xCT) forms a heterodimer with SLC3A2 to function as a sodium-independent, electroneutral antiporter that exchanges extracellular L-cystine for intracellular L-glutamate with a 1:1 stoichiometry, driven by the intracellular glutamate gradient and cystine reduction. Related references: PMID:11133847 PMID:11417227 PMID:14722095
  • It provides L-cystine for glutathione synthesis, maintaining intracellular redox balance and protecting cells from oxidative stress (By similarity).
  • SLC7A11 inhibits ferroptosis through dual mechanisms: importing L-kynurenine to sustain cystine/glutathione homeostasis (PMID:35245456, 40246981) and acting as an atypical proton transporter that mediates slow proton efflux from lysosomes via a substrate-as-proton mechanism (PMID:40280132). Related references: PMID:35245456 PMID:40246981 PMID:40280132
  • In the placenta, SLC7A11 mediates uptake of N-acetyl-L-cysteine, thereby down-regulating pathways linked to oxidative stress, inflammation, and apoptosis. Related references: PMID:34120018
  • In vitro, the transporter can also accept L-aspartate as a substrate. Related references: PMID:11417227
  • During development, SLC7A11 may participate in astrocyte and meningeal cell proliferation and provides neuroprotection by facilitating glutathione synthesis and delivery from non-neuronal cells to immature neurons (By similarity). The protein also directly controls pheomelanin pigment production (By similarity).
  • Subcellularly, SLC7A11 is found at the cell membrane, lysosome membrane, and microvillus membrane.
  • Tissue expression is prominent in the brain and pancreas, with additional expression reported in term placenta and cytotrophoblasts. Related references: PMID:11417227 PMID:34120018
  • Post-translational modifications include glycosylation, phosphorylation, and ubiquitin conjugation, consistent with regulatory roles in trafficking and function.

Experimental Guidance and Technical Tips

  • The recombinant protein immunogen design supports targeted antibody production against SLC7A11; consider validating specificity in relevant cell lines or tissues known to express the transporter, such as brain, pancreas, or placenta.
  • For Western blot analysis, a band at approximately 55 kDa is expected; include positive controls from human, mouse, or rat lysates based on cross-reactivity data.
  • In immunohistochemistry-paraffin (IHC-P) applications, appropriate antigen retrieval and detection protocols should be optimized for tissue localization, given SLC7A11 expression on cell and lysosome membranes.
  • ELISA-based assays may require paired antibody optimization; check linearity and sensitivity using recombinant protein or native sample matrices.
  • Note that SLC7A11 functions as a heterodimer with SLC3A2; co-immunoprecipitation or co-localization studies may require careful lysis and buffer conditions to preserve the interaction.
