Biological Background: SLC7A11/xCT Function and Localization
- SLC7A11 (xCT) forms a heterodimer with SLC3A2 to function as a sodium-independent, electroneutral antiporter that exchanges extracellular L-cystine for intracellular L-glutamate with a 1:1 stoichiometry, driven by the intracellular glutamate gradient and cystine reduction. - It provides L-cystine for glutathione synthesis, maintaining intracellular redox balance and protecting cells from oxidative stress (By similarity).
- SLC7A11 inhibits ferroptosis through dual mechanisms: importing L-kynurenine to sustain cystine/glutathione homeostasis (PMID:35245456, 40246981) and acting as an atypical proton transporter that mediates slow proton efflux from lysosomes via a substrate-as-proton mechanism (PMID:40280132).
