Biological Background: SLC6A2 Function and Localization
- SLC6A2, also known as the norepinephrine transporter (NET) or solute carrier family 6 member 2, is encoded by the SLC6A2 gene (alternative names: NAT1, NET1, SLC6A5). It undergoes alternative splicing, generating isoforms with potentially distinct functions.
- The protein mediates sodium- and chloride-dependent transport of norepinephrine, the primary neurotransmitter in the sympathetic nervous system, and is responsible for its reuptake and clearance from the synaptic cleft, thereby regulating neurotransmitter homeostasis. Related references: PMID:2008212, PMID:8125921, PMID:38750358
- In addition to norepinephrine, SLC6A2 can also transport dopamine in a sodium- and chloride-dependent manner, broadening its role in monoamine signaling. Related references: PMID:11093780, PMID:8125921, PMID:39395208
- Subcellularly, SLC6A2 is localized to the cell membrane, particularly at presynaptic axon terminals and synaptosomes, where it actively reclaims released neurotransmitters.
- The protein is characterized by the presence of disulfide bonds, glycosylation sites, lipidation (palmitoylation), and metal-binding motifs, which may contribute to its structural stability and functional regulation.
- SLC6A2 is a member of the neurotransmitter:sodium symporter family and contains multiple transmembrane helices characteristic of membrane transporters; it is annotated with keywords including alternative splicing, disease variant, glycoprotein, and synapse.
Experimental Guidance and Technical Tips
- The immunogen (N-terminal amino acids 1–70) may target an accessible epitope; consider confirming antibody specificity using SLC6A2-overexpressing cell models or tissue samples where the transporter is highly expressed.
- For Western blot (WB) analysis, the predicted molecular weight is ~69 kDa (UniProt: 69332 Da); run an appropriate molecular weight marker and consider potential post‑translational modifications that may shift the observed band.
- In ELISA applications, the antibody can be used for direct or sandwich formats; pairing with a polyclonal antibody or another monoclonal clone may improve sensitivity, and it is advisable to validate performance in the relevant sample matrix.
