Biological Background: SLC6A14 Function and Localization
- SLC6A14, also known as amino acid transporter ATB0+ or solute carrier family 6 member 14, is a member of the sodium- and chloride-dependent neurotransmitter transporter family.
- It functions as a broad-spectrum amino acid transporter, mediating the uptake of neutral and cationic amino acids (excluding proline) in a Na(+)/Cl(-)-dependent manner. Related references: PMID:10446133
- It transports non-alpha-amino acids such as beta-alanine with low affinity, and has a higher affinity for dipolar and cationic amino acids like leucine and lysine. Related references: PMID:18599538
- Additionally, SLC6A14 transports carnitine, butyrylcarnitine, and propionylcarnitine, coupling to transmembrane Na(+) and Cl(-) gradients. Related references: PMID:17855766
- The protein is localized at the apical cell membrane and plays a key role in intestinal amino acid absorption.
- Tissue expression is highest in adult and fetal lung, trachea, and salivary gland, with lower levels in mammary gland, stomach, pituitary gland, and very low levels in colon, uterus, prostate, and testis.
- Keywords from UniProt include amino-acid transport, cell membrane, glycoprotein, symport, and transmembrane helix, highlighting its membrane transport and glycosylation features.
- The predicted molecular weight is approximately 72 kDa, consistent with the supplied antibody specification.
Experimental Guidance and Technical Tips
- The immunogen used is a recombinant fusion protein covering amino acids 131-234 of human SLC6A14. This region is likely to be specific to the transporter and may be suitable for detecting endogenous SLC6A14 by western blot and ELISA.
- For WB analysis, consider using a positive control such as lysates from human lung or intestine, where SLC6A14 is abundantly expressed. Validate detection in the relevant sample system.
- The antibody cross-reacts with mouse SLC6A14, making it suitable for mouse model studies; however, sequence homology should be confirmed for other species if needed.
- Optimize antibody concentration and incubation conditions experimentally, as polyclonal antibodies may exhibit lot-to-lot variability.
- When performing ELISA, ensure proper coating and blocking conditions, and consider pairing with a suitable detection antibody if developing a sandwich assay.
