Biological Background: SLC5A2 Function and Localization
- SLC5A2 encodes the sodium/glucose cotransporter 2 (SGLT2), a member of the solute carrier family 5, functioning as a low-affinity electrogenic Na+/glucose symporter.

- SLC5A2 is predominantly expressed in the kidney, where it plays a critical role in reabsorbing glucose from the glomerular filtrate in the early proximal tubules.
- Subcellularly, SGLT2 localizes to the apical cell membrane of renal proximal tubular epithelial cells.
- The protein is a glycoprotein with multiple transmembrane helices and disulfide bonds, and alternative splicing generates multiple isoforms; mutations are linked to renal glucosuria.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the C-terminal region (amino acids 564-624) of human SLC5A2. Consider this when designing blocking peptides or competition assays.
- For Western blot, a band near 73 kDa is expected, but glycosylation may cause a shift; validate with relevant controls (e.g., deglycosylation enzymes) if needed.
- In immunohistochemistry (IHC-P), use FFPE kidney sections with appropriate antigen retrieval. For immunofluorescence (IF-P), frozen sections or cultured renal cell lines are recommended.
- Cross-reactivity with mouse and rat is indicated; however, always verify specificity in your sample system using positive and negative controls.
- For ELISA applications, standard coating and detection protocols can be adopted, but optimize antibody dilution individually.
