Biological Background: SLC5A1 Function and Localization
- SLC5A1 encodes the sodium/glucose cotransporter 1 (SGLT1), a high-affinity, Na+-dependent symporter of the solute carrier family 5.
- The protein mediates electrogenic co-transport of D-glucose or D-galactose with Na+ at a 2:1 stoichiometry, driven by the transmembrane Na+ gradient (PubMed:20980548, 34880492, 35077764). Related references: PMID:20980548, PMID:34880492, PMID:35077764
- In enterocytes, SGLT1 is the principal apical transporter for dietary glucose and galactose absorption; basolateral exit is mediated by GLUT2.
- Functions as a glucose sensor in enteroendocrine cells, triggering secretion of the incretins GIP and GCG to regulate appetite and energy homeostasis (PubMed:8563765). Related references: PMID:8563765
- In the kidney, SGLT1 participates in glucose reabsorption in the S3 segment of proximal tubules, complementing SGLT2-mediated reabsorption.
- Expressed in endometrial epithelial cells, where it imports glucose for glycogen synthesis and supports embryo nutritional needs and decidual transformation (PubMed:28974690). Related references: PMID:28974690
- Also functions as a passive water channel with aquaporin-like conductivity, contributing to transepithelial water flux in the small intestine.
- Localizes to the apical plasma membrane and is subject to glycosylation and phosphorylation; alternative splicing generates additional isoforms.
Experimental Guidance and Technical Tips
- The immunogen targets a synthetic peptide within the intracellular loop (aa 400-500); this region may be well-exposed for antibody binding.
- For western blotting, the predicted molecular weight is approximately 73 kDa; validate using lysates from human intestinal or endometrial tissue/cell lines.
- For ELISA, consider peptide blocking experiments to confirm signal specificity.
- Recommended applications include WB and ELISA based on supplier data; additional applications may require individual optimization.
- This product shows reactivity with human SLC5A1; cross-reactivity with other species should be verified experimentally.
- Shipment on ice bag indicates temperature sensitivity; store at recommended conditions upon receipt.
