  • SLC4A4, also known as NBCe1, kNBC1, or solute carrier family 4 member 4, is a multi-pass transmembrane glycoprotein and a member of the SLC4 bicarbonate transporter family.
  • Functions as an electrogenic sodium/bicarbonate cotransporter with a Na+:HCO3- stoichiometry of 1:2 or 1:3, playing a critical role in bicarbonate reabsorption and intracellular pH regulation.
  • Localizes predominantly to the basolateral membrane of polarized epithelial cells, including renal proximal tubule and pancreatic duct cells.
  • Highly expressed in kidney proximal tubules and pancreas; also detected in corneal endothelium, heart, skeletal muscle, liver, salivary glands, prostate, colon, stomach, thyroid, brain, and spinal cord.
  • Subject to alternative splicing, generating multiple isoforms (e.g., kNBC1, pNBC) with distinct N-terminal regions and tissue distributions.
  • Post-translational modifications include phosphorylation and glycosylation, which may influence transporter activity and membrane targeting.
  • Associated with human disease: mutations in SLC4A4 cause proximal renal tubular acidosis with ocular abnormalities and band keratopathy, and it is implicated in retinitis pigmentosa.
