Biological Background: SLC4A2 Function and Localization
- Encoded by the SLC4A2 gene, anion exchange protein 2 (AE2) is a multi-pass membrane protein also known as non-erythroid band 3-like protein (BND3L) and solute carrier family 4 member 2.
- Functions as a sodium-independent anion exchanger, mediating the electroneutral exchange of chloride for bicarbonate ions across the plasma membrane, thereby regulating intracellular pH and cell volume. Related references: PMID:15184086 PMID:34668226
- Plays a critical role in osteoclast differentiation and function, regulating bone resorption and calpain-dependent actin cytoskeleton organization through anion exchange-dependent pH control. Related references: PMID:34668226
- Essential for intracellular pH regulation in CD8+ T-cells upon CD3 stimulation, modulating CD8+ T-cell responses.
- Localizes primarily to the basolateral and apical cell membranes, consistent with its role in epithelial ion transport.
- Expressed in the liver, stomach, kidney, prostate, thyroid, and rectum, with splice variants exhibiting tissue-specific distribution.
- Subject to post-translational modifications including methylation and palmitoylation, and exists in several alternatively spliced isoforms.
- Anion transport protein with structural features resolved via 3D-structure determination.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 200–300 of human SLC4A2 (NP_001186623.1), a region that may confer target specificity; consider sequence alignment to verify uniqueness across splice variants.
- For Western blotting, SLC4A2 is predicted at ~137kDa; use appropriate positive (e.g., HEK293, HepG2) and negative controls, and validate band identity with knockdown or knockout samples if available.
- In ELISA, optimize coating concentration and blocking conditions to minimize background; polyclonal antibodies may exhibit lot-to-lot variation, so titrate each new batch.
- Although polyclonal antibodies offer high sensitivity, confirm specificity in relevant biological matrices and consider cross-reactivity with other SLC4 family members.
