Biological Background: SLC34A3 Function and Localization
- SLC34A3 encodes the sodium-dependent phosphate transport protein 2C (NaPi-2c), also known as Na(+)/Pi cotransporter 2C, a member of the solute carrier family 34 (SLC34) that mediates cellular phosphate uptake.
- The transporter actively cotransports phosphate with sodium ions at a 2:1 Na⁺:Pi stoichiometry, functioning as an electroneutral symporter in the renal brush border membrane.
Related references: PMID:11880379 - In humans, NaPi-2c is predominantly expressed in the kidney, localized to the apical membrane of proximal tubular epithelial cells, where it plays a critical role in phosphate homeostasis.
- Mutations in SLC34A3 are linked to hereditary hypophosphatemic rickets with hypercalciuria (HHRH), highlighting its clinical importance in renal phosphate wasting disorders.
- The protein undergoes glycosylation and phosphorylation and contains structural features such as disulfide bonds and multiple transmembrane helices, with a molecular weight of approximately 64 kDa.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (amino acids 1–117) of human SLC34A3; this antibody may recognize both full-length protein and N-terminal fragments in denaturing Western blots.
- For Western blotting, use kidney tissue lysates or SLC34A3-transfected cell lysates; expect a specific band around 64 kDa. Consider using reducing SDS-PAGE conditions to resolve disulfide-linked complexes.
- In ELISA, coat with recombinant SLC34A3 protein or a peptide encompassing the immunogen sequence, and titer the antibody to determine optimal working concentrations for your specific assay format.
- Cross-reactivity with mouse and rat SLC34A3 has been reported; validate the antibody in the relevant sample system, as detection may vary depending on sequence homology in the N-terminal region.
