Biological Background: SLC2A9 Function and Localization
- SLC2A9, also designated GLUT9 or URATv1, is a facilitative glucose transporter family member that primarily acts as a high-capacity urate transporter.
- Functional characterization demonstrates that SLC2A9 transports urate at rates 45–60 fold higher than glucose; it also exhibits residual low-affinity glucose and fructose transport, does not transport galactose, and mediates small adenine uptake. - The protein localizes predominantly to the cell membrane, with specific isoforms targeted to basolateral or apical membranes in polarized renal epithelial cells and placenta.
- Tissue expression is highest in kidney proximal tubules, liver, and placenta, with detection in lung, leukocytes, heart, skeletal muscle, and articular cartilage chondrocytes.
- Post-translational modifications include glycosylation and phosphorylation; alternative splicing generates multiple isoforms, and genetic variants are associated with hyperuricemia and gout.
