Biological Background: SLC29A3 Function and Localization
- SLC29A3 encodes equilibrative nucleoside transporter 3 (ENT3), a multi-pass membrane protein belonging to the solute carrier 29 family.
- It functions as a Na+-independent, facilitative uniporter for a broad range of nucleosides (adenosine, guanosine, uridine, thymidine, cytidine, inosine) and nucleobases, with transport activity enhanced at acidic pH. Related references: PMID:15701636, PMID:19164483, PMID:20595384, PMID:28729424
- Mediates export of nucleosides from lysosomes to cytosol and import into mitochondria, thereby facilitating nucleic acid salvage pathways. Related references: PMID:28729424
- Additionally capable of transporting monoamine neurotransmitters (dopamine, serotonin, noradrenaline, tyramine) and ATP. Related references: PMID:19164483
- Regulates macrophage function via lysosomal nucleoside export (By similarity).
- Acts as a lysosomal urate exporter and modulates inflammasome activation through its dual role in adenosine and urate transport. Related references: PMID:41476938
- Subcellularly localizes to lysosome membrane, late endosome membrane, mitochondrion membrane, and cell membrane.
- Widely expressed in adult and fetal tissues, with highest levels in placenta, uterus, ovary, spleen, lymph node, bone marrow, and liver; lower levels in brain and heart; also expressed in macrophages. Related references: PMID:15701636, PMID:19164483, PMID:22174130
Experimental Guidance and Technical Tips
- The immunogen spans the N‑terminal region (amino acids 1–110), which may be exposed in certain conformations; consider this when designing detection protocols for native versus denatured protein.
- For WB, the predicted molecular weight is approximately 52 kDa; validate with positive control lysates from tissues known to express high levels of SLC29A3, such as placenta or spleen.
- For ELISA, optimize coating conditions and confirm reactivity with recombinant or endogenous SLC29A3 in your sample system.
- Given the reported localization to lysosomes and mitochondria, subcellular fractionation or organelle enrichment may improve detection sensitivity in complex samples.
- Cross‑reactivity is reported for human; if working with other species, experimental validation is recommended.
