Biological Background: SLC27A2 Function and Localization
- Encodes long-chain fatty acid transport protein 2 (FATP2), a member of the solute carrier family 27 (SLC27A2). Also known as very long-chain acyl-CoA synthetase (VLACS).
- Facilitates import of long-chain fatty acids (LCFA) into cells and catalyzes their ATP-dependent conversion to fatty acyl-CoA, a key step preventing fatty acid efflux and driving uptake. Plays a pivotal role in regulating LCFA substrate availability for beta-oxidation and triglyceride synthesis in liver. Related references: PMID:10198260, PMID:10749848, PMID:11980911, PMID:20530735
- Activates very long-chain fatty acids and branched-chain fatty acids like phytanic acid and pristanic acid. Shows preference for n-3 fatty acids, generating CoA derivatives that are trafficked into phosphatidylinositol. Related references: PMID:10198260, PMID:21768100
- May contribute to synthesis of sphingosine-1-phosphate. Related references: PMID:24269233
- In vitro, activates THCA (a C27 bile acid precursor) but does not activate C24 bile acids; however, not critical for bile synthesis in vivo. Related references: PMID:11980911, PMID:20530735
- Located at endoplasmic reticulum membrane, peroxisome membrane, cell membrane, and microsome.

- Post-translational modifications include acetylation and phosphorylation; contains ATP-binding and transmembrane domains.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a central region (aa 290-573) of human FATP2; this region is conserved among species, consistent with observed cross-reactivity to mouse and rat.
- For Western blot, a band at approximately 70 kDa is expected; validate in the relevant sample system, such as liver lysate.
- In immunohistochemistry (paraffin), consider optimizing antigen retrieval and testing on liver or placenta sections.
- For immunofluorescence/ICC, permeabilization is required due to intracellular membrane localization; confirm with appropriate controls.
- Polyclonal antibodies may recognize multiple isoforms; verify specificity by using knockdown/knockout controls or other validation methods.
