Biological Background: SLC25A4/ANT1 Function and Localization
- SLC25A4 encodes ADP/ATP translocase 1 (ANT1, AAC1), a mitochondrial carrier protein also known as solute carrier family 25 member 4. It is predominantly expressed in heart and skeletal muscle.
- Functions as an ADP:ATP antiporter in the inner mitochondrial membrane, importing ADP for ATP synthesis and exporting ATP to fuel cellular processes (PubMed:21586654, PubMed:27693233, PubMed:23173940). Related references: PMID:21586654, PMID:27693233, PMID:23173940
- Operates via an alternating access mechanism with distinct cytoplasmic-open and matrix-open states (PubMed:37278158). Related references: PMID:37278158
- Also involved in mitochondrial uncoupling and regulation of the mitochondrial permeability transition pore (mPTP), contributing to cell death (PubMed:31883789). Related references: PMID:31883789
- Proton transporter activity requires free fatty acids and is inhibited by ADP:ATP antiporter activity, balancing ATP production and thermogenesis (By similarity).
- Independently regulates mitophagy through interaction with TIMM44, stabilizing PINK1 (By similarity).
- Localizes to the mitochondrial inner membrane (multi-pass membrane protein).
- Post-translational modifications include acetylation, phosphorylation, and S-nitrosylation; mutations cause primary mitochondrial diseases such as cardiomyopathy and progressive external ophthalmoplegia.
Experimental Guidance and Technical Tips
- For Western blot (WB), anticipate a specific band near 33 kDa. Use appropriate blocking and detection conditions.
- In immunofluorescence (IF/ICC), expect mitochondrial staining; co-localization with a mitochondrial marker is advisable.
- The immunogen (aa 40-298) is conserved across human, mouse, and rat; validate cross-reactivity in your specific sample system.
- ELISA applications may be optimized with recombinant or purified protein.
