Biological Background: SLC25A24 Function and Localization

  • SLC25A24 encodes the mitochondrial adenyl nucleotide antiporter SLC25A24, also known as SCaMC-1, APC1, or solute carrier family 25 member 24.
  • Acts as an electroneutral antiporter that mediates transport of adenyl nucleotides through the inner mitochondrial membrane; originally identified as an ATP-Mg/Pi antiporter but with broad specificity for adenyl nucleotides. Related references: PMID:15123600 PMID:22015608
  • Regulates the mitochondrial matrix adenyl nucleotide pool, adapting to cellular energetic demands and indirectly modulating nucleotide-dependent metabolic pathways. Related references: PMID:15123600 PMID:22015608
  • In vitro, exhibits low activity with guanyl and pyrimidine nucleotides. Related references: PMID:15123600
  • May protect cells against oxidative stress-induced cell death by buffering mitochondrial calcium levels via formation of calcium-phosphate precipitates. Related references: PMID:22015608 PMID:29100093
  • Localizes to the mitochondrial inner membrane.
  • Expressed ubiquitously, with highest levels in testis, intermediate in small intestine and pancreas, and weak in kidney, spleen, liver, skeletal muscle, and heart.
  • Features alternative splicing and acetylation; contains mitochondrial carrier domains and transmembrane helices; a 3D structure is available.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 1–200 (N-terminal region) of human SLC25A24; the antibody may recognize this epitope region. Consider using the recombinant immunogen as a positive control in ELISA.
  • For Western blotting (WB), a band around 53 kDa is expected. Validate with mitochondrial-enriched fractions or lysates from high-expression tissues (e.g., testis).
  • In immunofluorescence/immunocytochemistry (IF/ICC), colocalization with mitochondrial markers is recommended to confirm specificity. Cell permeabilization and fixation conditions should be optimized for mitochondrial antigens.
  • This polyclonal antibody has reported cross-reactivity with human, mouse, and rat SLC25A24; validate reactivity in the species and sample system relevant to your experiments.
