Biological Background: SLC25A20 Function and Localization
- SLC25A20, also known as carnitine/acylcarnitine translocase (CACT), is a mitochondrial carrier protein belonging to the solute carrier family 25.
- Mediates the electroneutral exchange of acylcarnitines of various chain lengths with free carnitine across the mitochondrial inner membrane via a ping-pong mechanism, a critical step in mitochondrial fatty acid oxidation. Related references: PMID:12892634 PMID:18307102
- Also catalyzes unidirectional transport (uniport) of carnitine, albeit at lower rates than the exchange reaction. Related references: PMID:18307102
- Plays a key role in the carnitine shuttle by delivering acylcarnitines into the mitochondrial matrix where carnitine palmitoyltransferase 2 (CPT-2) releases them for beta-oxidation.
- Localizes to the mitochondrial inner membrane, consistent with its transport function.
- Post-translational modification includes acetylation, and the protein is linked to disease variants (UniProt keyword 'Disease variant'), highlighting its clinical relevance in metabolic disorders.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide corresponding to amino acids 1–100 of human SLC25A20, thus the antibody may recognize epitopes in the N-terminal region; consider verifying specificity with peptide blocking.
- In Western blot, a specific band around 33 kDa is expected; validate detection using mitochondrial-enriched lysates from human, mouse, or rat tissues as positive controls.
- For ELISA, both direct and indirect formats can be explored; using the immunizing peptide as a competitor may help confirm specific binding.
- Since cross-reactivity is reported for human, mouse, and rat, pre-test the antibody in your model system to ensure consistent performance across species.
