Biological Background: SLC25A13/Citrin Function and Localization
- SLC25A13, also known as citrin or Aralar2, is an electrogenic aspartate/glutamate antiporter located in the mitochondrial inner membrane.
- It plays a key role in the malate-aspartate shuttle, mediating the efflux of aspartate and the coupled uptake of glutamate and a proton into mitochondria. Related references: PMID:11566871, PMID:38937634, PMID:38945283
- The transporter can also facilitate the uptake of L-cysteinesulfinate in exchange for L-glutamate and a proton, or aspartate without proton translocation. Related references: PMID:11566871
- It does not transport gamma-aminobutyric acid (GABA). Related references: PMID:38945283
- SLC25A13 is highly expressed in the liver, with lower levels in the kidney, pancreas, placenta, heart, and brain.
- The protein is involved in calcium binding and is associated with intrahepatic cholestasis (citrin deficiency) as a disease variant.
- Post-translational modifications include acetylation, methylation, and phosphorylation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1–300) of human SLC25A13. For WB, consider using the predicted molecular weight of 74 kDa as a reference.
- For IHC-P, validate in tissues with high expression such as liver; antigen retrieval conditions may need optimization.
- Cross-reactivity with mouse and rat suggests conserved epitopes; verify performance in these species experimentally.
- Due to the mitochondrial localization, ensure appropriate lysis and sample preparation for WB or ELISA to expose the membrane protein.
