Biological Background: SLC25A13 Function and Localization
- SLC25A13, also known as citrin or ARALAR2, is a mitochondrial inner membrane protein belonging to the solute carrier family 25. It is highly expressed in the liver, with lower levels in kidney, pancreas, placenta, heart, and brain.
- It functions as an electrogenic aspartate/glutamate antiporter, essential for the malate-aspartate shuttle, catalyzing the exchange of mitochondrial aspartate for cytosolic glutamate and a proton. It also transports L-cysteinesulfinate. Related references: PMID:11566871 PMID:38937634 PMID:38945283
- The protein contains calcium-binding motifs and is regulated by calcium levels, linking mitochondrial metabolism to cellular signaling.
- Mutations in SLC25A13 are associated with citrullinemia type II (CTLN2) and neonatal intrahepatic cholestasis caused by citrin deficiency (NICCD), highlighting its clinical importance.
- The protein undergoes post-translational modifications including acetylation, methylation, and phosphorylation, which may modulate its activity.
- SLC25A13 is localized to the mitochondrion inner membrane and functions as an antiporter, consistent with its role in metabolite exchange across the membrane.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (amino acids 1-300) of human SLC25A13, which may contribute to recognition of both human and rodent forms, as indicated by cross-reactivity with mouse and rat samples.
- For Western blot, a band at approximately 74 kDa is expected; ensure appropriate gel percentage and transfer conditions for this size.
- Consider optimizing antibody dilution for ELISA and WB based on target abundance in your specific sample type, especially given the variable tissue expression (high in liver).
- Since SLC25A13 is a mitochondrial membrane protein, sample preparation may require mitochondrial enrichment or use of detergents to fully solubilize the protein.
- Validate antibody performance in relevant model systems, such as liver tissue or cell lines, where expression is highest.
