Biological Background: SLC25A11 Function and Localization
- SLC25A11 (Solute carrier family 25 member 11), also known as 2-oxoglutarate/malate carrier (OGC) or SLC20A4, is a mitochondrial carrier protein belonging to the SLC25 family. It is encoded by the SLC25A11 gene, which gives rise to multiple isoforms through alternative splicing.
- SLC25A11 catalyzes the electroneutral exchange of 2-oxoglutarate (alpha-oxoglutarate) for malate across the inner mitochondrial membrane, a key step in several metabolic shuttles. Related references: PMID:25637873 PMID:38937634
- It can also transport other dicarboxylic acids such as malonate, succinate, maleate, and oxaloacetate with lower affinity, but does not transport glutathione.
- The transport mechanism proceeds via a consecutive exchange model, where one substrate is fully translocated before the second substrate binds for counter-transport. Related references: PMID:38937634
- In addition to its antiport activity, SLC25A11 can facilitate proton transport in the presence of long-chain fatty acid protonophores, contributing to mitochondrial uncoupling and regulation of energetic efficiency.

Experimental Guidance and Technical Tips
- This rabbit polyclonal antibody was raised against a synthetic peptide derived from the N-terminal region (amino acids 1–100) of human SLC25A11. It is predicted to recognize endogenous SLC25A11 in human, mouse, and rat samples.
- For Western blot (WB), a band at approximately 34 kDa is expected. Due to post-translational modifications or alternative splicing, additional bands may appear; validate the expected molecular weight in your system.
- For IHC-P and IF/ICC, consider including appropriate controls (e.g., peptide blocking or knockdown samples) to confirm specificity. Optimal dilution and incubation conditions should be determined empirically.
- Polyclonal antibodies may exhibit lot-to-lot variability; perform a pilot test when switching to a new lot.
