Biological Background: SLC23A2 Function and Localization
- SLC23A2 encodes the Na(+)/L-ascorbic acid transporter 2 (SVCT2), a member of the solute carrier family 23, also known as nucleobase transporter-like 1 protein (NBTL1) and yolk sac permease-like molecule 2 (YSPL2).
- Functions as a sodium/ascorbate cotransporter that mediates high-affinity, electrogenic uptake of vitamin C with a stoichiometry of 2 Na+ for each ascorbate molecule. Related references: PMID:10471399 PMID:10556521
- Ubiquitously expressed across human tissues, playing a central role in maintaining intracellular ascorbate levels for antioxidant protection and enzymatic cofactor functions.
- Localizes predominantly to the cell membrane, consistent with its role as a plasma membrane transporter.
- Contains multiple transmembrane domains and is subject to post-translational modifications including glycosylation and phosphorylation; calcium-binding regions are also predicted.
- Alternative splicing generates distinct isoforms, which may exhibit tissue-specific regulation or functional diversity.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide corresponding to the C-terminal region (amino acids 551–650) of human SLC23A2; this region may be conserved across splice variants, enabling broad isoform detection.
- For Western blot, the predicted molecular weight is approximately 70 kDa. Due to glycosylation, the observed band may shift slightly. Use reducing conditions and validate with positive controls such as human or mouse tissue lysates known to express SVCT2.
- For ELISA, consider optimizing coating antigen and antibody dilution using recombinant protein or peptide. Include appropriate negative controls to assess specificity.
- Given the membrane localization, enrichment of membrane protein fractions may enhance detection sensitivity in some sample types.
- Cross-reactivity with mouse is reported; validate in the relevant experimental system when switching species.
