Biological Background: SLC1A7 Function and Localization
- Excitatory amino acid transporter 5 (EAAT5), also known as solute carrier family 1 member 7 (SLC1A7) or retinal glutamate transporter, is encoded by the SLC1A7 gene in humans.
- Functions as a sodium-dependent, high-affinity amino acid transporter mediating the uptake of L-glutamate, L-aspartate, and D-aspartate; acts as a symporter with 2-3 Na+ ions and one proton, counter-transporting K+. Related references: PMID:9108121
- Additionally serves as an inhibitory glutamate-gated chloride channel, mediating a large Cl- conductance that affects visual processing at mammalian rod bipolar cell axon terminals (by similarity).
- Localizes to the photoreceptor inner segment membrane and synaptic cell membrane, consistent with its role in retinal signaling.
- Predominantly expressed in the retina, with detectable levels also in liver, heart, muscle, and brain, implying potential extra-retinal functions.
- Subject to alternative splicing and glycosylation, which may regulate its transport activity or subcellular distribution.
Experimental Guidance and Technical Tips
- The immunogen targets amino acids 480-560 within the C-terminal region of human SLC1A7; consider verifying isoform specificity in your experimental system.
- For Western blot, a band around 61 kDa is expected; validate with appropriate positive and negative controls to confirm specificity.
- Reported mouse cross-reactivity suggests utility in rodent models; always validate in your specific tissue or cell line.
- ELISA applications may require optimization of coating antigen and antibody concentration; include relevant controls for accurate quantification.
