Biological Background: SLC1A5 Function and Localization
- SLC1A5, also known as Neutral amino acid transporter B(0) or ASCT2, is a sodium-coupled neutral amino acid antiporter that mediates the bidirectional exchange of amino acids, with L-glutamine as the preferred substrate, coupling its transport to at least one sodium ion. Related references: PMID:17094966; PMID:23756778; PMID:26492990
- Through L-glutamine uptake, SLC1A5 fuels biosynthesis of proteins, nucleotides, and amine sugars and supports mitochondrial oxidative phosphorylation, driving the metabolic switch toward cell differentiation in stem and activated cells. Related references: PMID:23756778; PMID:24953180
- It transports extracellular L-glutamate in a pH-dependent manner, co-transporting one proton and one sodium ion in exchange for intracellular L-glutamine, thereby participating in the glutamine/glutamate cycle in the nervous system. Related references: PMID:32733894
- SLC1A5 mediates the release and reabsorption of D-serine from retinal glia and renal proximal tubules, respectively, modulating NMDA receptor function and D-serine homeostasis. Related references: PMID:17094966; PMID:38650461
- The transporter also displays an uncoupled, sodium- and amino acid-dependent channel-like anion conductance with a preference for SCN(-), NO3(-), I(-), and Cl(-).
- Beyond its transport function, SLC1A5 acts as a cell surface receptor for several retroviruses, including feline endogenous virus RD114, baboon M7 virus, and type D simian retroviruses, and mediates syncytin-1-induced trophoblast fusion in placental development. Related references: PMID:10708449; PMID:23492904; PMID:38671230
- Subcellularly, it is found at the cell membrane and apical membrane, and is also identified in melanosomes, with tissue expression in placenta, lung, skeletal muscle, kidney, pancreas, intestine, and CD34+ hematopoietic progenitors. Related references: PMID:8702519; PMID:24953180
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 154–224 within the central region of human SLC1A5, which may favor recognition of the native topology if epitopes are accessible; consider evaluating binding in both denaturing and native conditions if needed.
- For Western blotting, the predicted molecular weight is approximately 57 kDa; due to glycosylation and possible dimerization, observed bands may shift; validate using positive control lysates from placenta or lung tissues.
- The reported cross-reactivity with human, mouse, and rat suggests conservation of the epitope region; confirm reactivity in your specific experimental model by running a species-matched positive control.
- ELISA applications may require optimization of coating antigen and antibody concentration; consider using the recombinant immunogen as a standard.
- As a rabbit polyclonal antibody, batch-to-batch variability is possible; include internal reference controls and re-validate when switching to a new lot.
