Biological Background: SLC1A4 Function and Localization
- SLC1A4 encodes the neutral amino acid transporter A (ASCT1), also known as alanine/serine/cysteine/threonine transporter 1, a member of the solute carrier family 1.
- Functions as a sodium-coupled antiporter that exchanges neutral amino acids such as L- and D-serine, L-threonine, L-asparagine, and L-alanine across the plasma membrane. Related references: PMID:14502423, PMID:26041762, PMID:27272177
- Plays a critical role in D-serine homeostasis in astrocytes at excitatory synapses, mediating electrogenic D-serine influx coupled to L-serine efflux and supporting de novo D-serine synthesis in neurons.
- Displays a sodium- and amino acid-dependent but uncoupled channel-like anion conductance with a preference SCN(-) > NO3(-) > I(-) > Cl(-). Related references: PMID:8910405
- Localizes to the cell membrane and melanosome membrane.
- Expressed in most tissues, with highest levels in brain, muscle, and pancreas.
- Post-translational modifications include acetylation, phosphorylation, and glycosylation; alternative splicing generates multiple isoforms.
- Genetic variants are associated with neurological disorders including epilepsy and intellectual disability.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 140–220, a conserved cytoplasmic region, supporting the demonstrated cross-reactivity with human, mouse, and rat.
- For Western blot, expect a band of approximately 56 kDa; consider using positive control lysates from brain or pancreas tissues.
- Validate antibody performance in your specific sample and application, as optimal working dilutions may vary.
