Biological Background: SLC1A1/EAAT3 Function and Localization
- SLC1A1 encodes the excitatory amino acid transporter 3 (EAAT3), also known as EAAC1 and hEAAC1, a member of the solute carrier family 1 (SLC1) of high-affinity sodium-dependent glutamate transporters.
- The protein functions as a symporter that mediates the uptake of L-glutamate, L-aspartate, and D-aspartate, coupled to the co-transport of Na+ and H+ ions and the counter-transport of K+ ions, essential for maintaining extracellular glutamate homeostasis. Related references: PMID:21123949, PMID:26690923, PMID:33658209, PMID:7521911
- It can also transport L-cysteine, contributing to glutathione biosynthesis and cellular protection against oxidative stress. Related references: PMID:21123949
- Additionally, EAAT3 mediates a chloride flux that is not coupled to amino acid transport, which helps avoid the accumulation of negative charges during symport activity. Related references: PMID:26690923, PMID:8857541
- EAAT3 is primarily localized at the cell membrane, including the apical surface of epithelial cells, as well as at synaptic terminals, early, late, and recycling endosomes.
- It is widely expressed in tissues such as liver, muscle, testis, ovary, and brain, with dense expression in the substantia nigra, red nucleus, hippocampus, and cerebral cortex.
- The transporter plays a critical role in L-glutamate and L-aspartate reabsorption in renal tubules. Related references: PMID:21123949
- Post-translational modifications include phosphorylation and glycosylation, as indicated by UniProt keywords; genetic variations in SLC1A1 are associated with schizophrenia.
Experimental Guidance and Technical Tips
- The immunogen was designed against the C-terminal region (amino acids 430–524) of human SLC1A1, which may recognize epitopes conserved across human, mouse, and rat, consistent with the observed cross-reactivity.
- For Western blotting, consider the predicted molecular weight of 57 kDa and validate the antibody in relevant tissue lysates such as brain or kidney.
- In immunofluorescence (IF-P), permeabilization may be necessary to detect intracellular pools, and signal could be observed at the plasma membrane and endosomal compartments.
- ELISA performance should be verified using the specific coating antigen or recombinant protein to ensure sensitivity and specificity.
- As a polyclonal antibody, lot-to-lot consistency is typical; users are encouraged to include appropriate positive and negative controls in each experiment.
