Biological Background: Urea Transporter 1 Function and Localization
- SLC14A1 encodes urea transporter 1 (UT-B), a member of the solute carrier family 14, also known as the Kidd (JK) blood group antigen.
- Mediates the transport of urea driven by a concentration gradient across the cell membrane of erythrocytes (PubMed:10514515, PubMed:7797558, PubMed:7989337). Related references: PMID:10514515, PMID:7797558, PMID:7989337
- Also mediates urea transport across the basolateral membrane of renal inner medullary collecting duct cells, playing a critical role in the urinary concentrating mechanism (By similarity).
- Facilitates water transport in erythrocytes (By similarity).
- Subcellular localization: predominantly expressed on the cell membrane, with basolateral polarization in renal epithelia.
- Detected in erythrocytes at the protein level (PubMed:23219802) and expressed in spleen erythroblasts and tumoral kidney (PubMed:7989337). Related references: PMID:23219802, PMID:7989337
- Post-translational modifications include glycosylation; the protein spans the membrane multiple times and exhibits alternative splicing, giving rise to several isoforms.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a synthetic peptide from the intracellular C-terminal region (amino acids 289–389). The antibody is expected to recognize an epitope that is not exposed on the cell surface, which may be relevant for applications requiring permeabilization.
- For Western blotting, a band around 43 kDa is anticipated; note that glycosylation may cause slight migration shifts. Optimize blocking and antibody dilution for your specific sample lysate.
- In IHC-P and IF/ICC, expect membrane staining. For intracellular epitope detection, permeabilization is required. Validate in positive control tissues such as kidney and erythroid lineage samples.
- For ELISA, consider using the recombinant extracellular domain or a peptide standard, and evaluate performance in sandwich pair setups.
