Biological Background: SLAMF7 Function and Localization

  • SLAMF7 is a self-ligand receptor of the SLAM family, also known as CD2 subset 1, CD2-like receptor-activating cytotoxic cells (CRACC), Membrane protein FOAP-12, Novel Ly9, Protein 19A, and CD319.
  • Isoform 1 (canonical) mediates NK cell activation via a SH2D1A-independent ERK pathway and positively regulates NK cell functions downstream of SH2D1B, involving PLCG1, PLCG2, and PI3K. Related references: PMID:11698418 PMID:16339536
  • In the absence of SH2D1B, isoform 1 inhibits NK cell function; it also acts inhibitory in T-cells.
  • Isoform 3 does not mediate NK cell activation and is expressed at much lower levels than isoform 1.
  • SLAMF7 may contribute to lymphocyte adhesion. Related references: PMID:11802771
  • In LPS-activated monocytes, SLAMF7 negatively regulates production of pro-inflammatory cytokines. Related references: PMID:23695528

  • The immunogen spans the extracellular region, suggesting the antibody may detect the receptor in native form; non-reducing conditions for ELISA may be considered.
  • Cross-reactivity with mouse is listed; validate in mouse primary immune cells or tissues such as spleen.
  • Due to alternative splicing, ensure isoform-specific detection if needed; validate with appropriate controls.
