Biological Background: SLAMF7 Function and Localization
- SLAMF7 (also known as CS1, CD319, CRACC) is a transmembrane receptor of the signaling lymphocytic activation molecule (SLAM) family, with two main isoforms produced by alternative splicing; isoform 1 mediates immune cell activation, while isoform 3 lacks activating function.
- Isoform 1 triggers NK cell activation through an SH2D1A-independent ERK-mediated pathway and positively regulates NK cell functions via phosphorylated SH2D1B, involving downstream signaling through PLCG1, PLCG2, and PI3K. Related references: PMID:11698418, PMID:16339536
- In the absence of SH2D1B, SLAMF7 can inhibit NK cell function; it also plays an inhibitory role in T-cells. Additionally, in LPS-activated monocytes, it negatively regulates the production of pro-inflammatory cytokines. Related references: PMID:23695528
- SLAMF7 may contribute to lymphocyte adhesion. Related references: PMID:11802771
- It is a membrane glycoprotein containing an immunoglobulin domain, and is classified under both innate and adaptive immunity keywords.
- SLAMF7 is predominantly expressed in immune-related tissues including spleen, lymph node, peripheral blood leukocytes, and bone marrow, with high levels in NK cells, activated B-cells, and monocytes. Isoform 3 is expressed at much lower levels.
- As a glycosylated protein, its observed molecular weight may differ from the predicted 37 kDa.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 23–226 of human SLAMF7 (extracellular domain), which may enable detection of both membrane-bound and soluble receptor forms.
- Due to glycosylation, the apparent molecular weight in Western blot may exceed the predicted 37 kDa; validate the band size using appropriate controls.
- This polyclonal antibody is suitable for WB and ELISA; optimize dilution, blocking, and incubation conditions for your specific sample type.
- Cross-reactivity with human, mouse, and rat SLAMF7 allows use in common experimental models; confirm specificity by testing in relevant tissues or cell lines.
