Biological Background: SKP2 Function and Localization
- SKP2 (S-phase kinase-associated protein 2), also known as p45, FBL1, FBXL1, is a substrate recognition component of the SCF (SKP1-CUL1-F-box protein) E3 ubiquitin ligase complex that mediates ubiquitination and proteasomal degradation of target proteins involved in cell cycle progression, signal transduction, and transcription. - SKP2 specifically recognizes phosphorylated CDKN1B/p27kip, promoting its degradation to regulate G1/S transition; this process also requires the cofactor CKS1.
- It targets a wide array of substrates, including ORC1, CDT1, RBL2, KMT2A/MLL1, CDK9, RAG2, NBN, FOXO1, UBP43, YTHDF2, and likely MYC, TOB1, and TAL1. Degradation of TAL1 requires cooperation with the E3 ligase STUB1. Related references: PMID:11931757, PMID:12435635, PMID:12769844, PMID:17962192

- Upon DNA damage, phosphorylated SKP2 mediates Lys-63-linked ubiquitination of NBN, facilitating ATM recruitment and promoting homologous recombination repair. Related references: PMID:22464731

- The protein localizes to both the cytoplasm and nucleus and is subject to post-translational modifications including acetylation and phosphorylation; it contains leucine-rich repeats characteristic of F-box proteins.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 100–200 of human SKP2; confirm that this region is sufficient for detecting the target in your specific application and sample type.
- For Western blot, consider a starting dilution range of 1:500–1:2000, and optimize based on expression levels in your model. A band near 48 kDa is expected.
- In immunofluorescence/ICC, include appropriate fixation and permeabilization steps to detect SKP2 in both cytoplasmic and nuclear compartments.
- For ELISA, evaluate compatibility with your coating buffer and detection system, and consider using this antibody as either capture or detection reagent after pairing validation.
- This antibody is raised against human SKP2; cross-reactivity with other species has not been determined. Validate specificity if used with non-human samples.
