Biological Background: SKP2 Function and Localization

  • SKP2 (S-phase kinase-associated protein 2), also known as p45, FBL1, FBXL1, is a substrate recognition component of the SCF (SKP1-CUL1-F-box protein) E3 ubiquitin ligase complex that mediates ubiquitination and proteasomal degradation of target proteins involved in cell cycle progression, signal transduction, and transcription.   • SKP2 specifically recognizes phosphorylated CDKN1B/p27kip, promoting its degradation to regulate G1/S transition; this process also requires the cofactor CKS1.
  • It targets a wide array of substrates, including ORC1, CDT1, RBL2, KMT2A/MLL1, CDK9, RAG2, NBN, FOXO1, UBP43, YTHDF2, and likely MYC, TOB1, and TAL1. Degradation of TAL1 requires cooperation with the E3 ligase STUB1. Related references: PMID:11931757, PMID:12435635, PMID:12769844, PMID:17962192
