Biological Background: SKP2 Function and Localization
- SKP2 (S-phase kinase-associated protein 2) is an F-box protein that serves as the substrate recognition subunit of the SCF (SKP1-CUL1-F-box protein) E3 ubiquitin ligase complex. It targets key cell cycle regulators, such as CDKN1B/p27 and CDKN1A, for ubiquitination and proteasomal degradation, thereby promoting G1/S transition (PubMed:9736735, PubMed:11931757, PubMed:12435635). Related references: PMID:9736735, PMID:11931757, PMID:12435635
- Beyond cell cycle control, SKP2 mediates ubiquitination of diverse substrates including CDT1, ORC1, FOXO1, MYC, and CDH1, influencing DNA replication, apoptosis, transcription, and cell migration (PubMed:12769844, PubMed:22770219). Related references: PMID:12769844, PMID:22770219
- In response to DNA damage, phosphorylated SKP2 catalyzes Lys-63-linked ubiquitination of NBN, facilitating ATM recruitment and homologous recombination repair (PubMed:22464731). Related references: PMID:22464731
- SKP2 also possesses antiviral activity by targeting hepatitis C virus non-structural protein 5A for proteasomal degradation, linking it to innate immunity.
- Localized to both the nucleus and cytoplasm, SKP2 undergoes post-translational modifications including phosphorylation and acetylation, which modulate its stability and function.
- SKP2 is a member of the F-box/LRR-repeat protein family (FBXL1) and is frequently overexpressed in various cancers, making it a potential diagnostic and therapeutic target.
Experimental Guidance and Technical Tips
- The antibody was raised against a synthetic peptide from the N-terminal region (aa 1-100) of human SKP2. For Western blotting, a band at approximately 48 kDa is expected in human cell lysates; consider using a reducing gel and validate with a positive control such as HeLa or 293T cells.
- For immunohistochemistry (paraffin-embedded tissue), antigen retrieval methods and antibody dilution require optimization; include appropriate tissue controls known to express SKP2 (e.g., tonsil, breast carcinoma).
- In ELISA applications, use the recombinant antibody as a capture or detection reagent in combination with validated antigen or matched antibody pairs; confirm performance with recombinant SKP2 protein.
- Cross-reactivity with other species has not been reported; this product is intended for human samples. Researchers should independently verify specificity in their model system.
