Biological Background: SKIV2L2 (MTR4) Function and Localization
- SKIV2L2, also known as MTR4, DOB1, or KIAA0052, is an ATP-dependent RNA helicase encoded by the MTREX gene in humans. It catalyzes ATP-dependent unwinding of RNA duplexes with a single-stranded 3' RNA extension.
Related references: PMID:27871484 PMID:29844170 PMID:29906447 - SKIV2L2 serves as a central subunit of the nuclear exosome targeting (NEXT), poly(A) tail exosome targeting (PAXT), and TRAMP-like complexes, directing subsets of non-coding and aberrant transcripts for exosomal degradation.
Related references: PMID:21855801 PMID:27871484 PMID:29844170 - It is involved in the 3'-processing of 7S pre-rRNA to mature 5.8S rRNA within the RNA exosome complex.
Related references: PMID:17412707 PMID:29107693 - SKIV2L2 may participate in pre-mRNA splicing and is a component of the spliceosome.
- It can unwind DNA:RNA heteroduplexes and displace DNA strands while translocating on RNA, facilitating the degradation of structured RNA substrates.
Related references: PMID:29844170 PMID:29906447 - The protein is localized to the nucleoplasm, nucleolus, and nuclear speckles.
- Post-translational modifications include acetylation and phosphorylation, with an isopeptide bond formation.
- SKIV2L2 also plays a role in the DNA damage response.

Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 1-304 of human SKIV2L2 (N-terminal region), which may aid in detecting the full-length protein in Western blotting.
- For Western blot (WB), the predicted molecular weight is approximately 118 kDa; ensure appropriate gel percentage and transfer conditions.
- In immunohistochemistry (IHC-P), antigen retrieval and antibody titration are recommended to optimize staining specificity.
- For immunoprecipitation (IP), use appropriate controls, such as non-immune IgG, to confirm specific interactions.
- The antibody cross-reacts with human, mouse, and rat SKIV2L2, making it suitable for studies across these species, but validation in the target species' tissue or cell type is advised.
