Biological Background: SIX5 Function and Localization
- SIX5 (also known as BOR2, DMAHP) is a homeobox transcription factor belonging to the SIX family, characterized by a homeodomain DNA-binding region.
- It functions as a transcriptional activator, binding to specific DNA motifs such as the ARE element (5'-GGTGTCAG-3') in ATP1A1 and the MEF3 element (5'-TCA[AG][AG]TTNC-3') in the myogenin and IGFBP5 promoters.
- SIX5 is involved in regulating organogenesis, with a proposed role in the determination and maintenance of retina formation.
- This protein interacts with Dach and Eya family members; its transcriptional activity can be coactivated by EYA1, EYA2, and EYA3.
- Subcellular localization includes both the nucleus and cytoplasm, consistent with a transcription factor that may shuttle between compartments.
- In adults, SIX5 is expressed in the eye, including corneal epithelium and endothelium, lens epithelium, ciliary body epithelia, retina layers, and sclera; it is not detected in fetal eyes.
- UniProt annotations highlight its roles in development, deafness-associated disease variants, and DNA-binding-dependent transcription regulation.
Experimental Guidance and Technical Tips
- The immunogen (aa 1-170) targets the N-terminal region of SIX5; this region lies outside the homeodomain and may enable detection of full-length endogenous protein.
- For Western blot, a band near 75 kDa is expected; inclusion of appropriate loading controls and molecular weight markers is advised.
- Given the reported expression in adult eye tissues, lysates from retinal or corneal samples may serve as positive controls for human, mouse, or rat.
- Cross-reactivity with mouse and rat SIX5 is indicated; however, experimental validation in the relevant sample system is recommended.
- As a polyclonal antibody, consider lot-to-lot consistency and possible nonspecific bands; titration and blocking optimization may be necessary.
- For ELISA, the antibody can be used as a capture or detection reagent, but optimal dilutions must be determined empirically.
