Biological Background: SIX2 Function and Localization

  • Homeobox protein SIX2 is a transcription factor that plays a critical role in the development of multiple organs, including kidney, craniofacial structures, stomach, and potentially limbs (by similarity).
  • During kidney development, SIX2 maintains cap mesenchyme nephron progenitor cells in an undifferentiated state by opposing ureteric bud signals, and cooperates with WNT9B to promote progenitor proliferation. It interacts with TCF7L2 and OSR1 in a Wnt-independent manner to repress differentiation genes such as WNT4, while also activating cap mesenchyme genes including itself, GDNF, and OSR1.
  • In craniofacial development, SIX2 governs elongation of the cranial base through regulation of chondrocyte differentiation. In stomach organogenesis, it controls pyloric sphincter formation and mucosal growth via a gene network including NKX2-5, BMPR1B, BMP4, SOX9, and GREM1.
  • SIX2 localizes to the nucleus and contains a homeobox DNA-binding domain, consistent with its role as a developmental regulator. It is classified under keywords: Developmental protein, DNA-binding, Homeobox, Nucleus, and Reference proteome.
  • At the protein level, SIX2 is strongly expressed in skeletal muscle and is detected in Wilms' tumor and fetal kidney cap mesenchyme, reflecting its role in kidney development and oncogenic contexts.
  • Additional functions include mediating HOXA2 control over the insulin-like growth factor pathway during branchial arch development, and a role in cell proliferation and migration (by similarity).

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 120–174 of human SIX2 (NP_058628.3); this region may be conserved across species, supporting the observed cross-reactivity with rat.
  • For Western blot, consider using nuclear-enriched lysates and validate the observed band against the predicted molecular weight of ~32 kDa. Endogenous SIX2 expression is tissue-specific, so include appropriate positive controls (e.g., skeletal muscle or fetal kidney samples).
  • For ELISA, the polyclonal nature may provide high sensitivity; however, titration experiments are recommended to determine optimal antibody and antigen concentrations.
  • Cross-reactivity has been reported with human and rat; if using other species, verify sequence homology in the immunogen region.
